Application of Electrophoresis in Testing the Genetic Purity of F1 Hybrid Seeds of Radish (Raphanus sativus)


Abstract
Crude extracts from single cotyledons of 2- to 3-days old seedlings of radish were separated by starch gel electrophoresis and stained for malate dehydrogenase (MDH) and acid phosphatase (AcPH) to investigate the possibility of utilizing isozyme variation in determining the rate of genetic impurity caused by mainly sib or/and selfing during the production of F©û hybrid seeds.
The varietal differences in isozyme banding pattern were found in the end of anodal zone for MDH (MDH-1) and in the cathodol zone for AcPH (AcPH-1), which suggests that this isozyme analysis can be used in testing the rate of contamination in radish hybrid varieties. Data from parental inbreds and their F©û¢¥s, F©ü¢¥s and backcross progenies revealed that the bands in both MDH-1 and AcPH-1 zones were controlled by two co-dominant alleles at respective single locus. While the banding pattern of MDH-1 was not changed with the age of the seedlings, that of AcPH-1 showed a progressive reduction in the number of bands as well as in its activity as the seedlings became older. However, both enzyme can serve the purpose of testing the impurity of F©û hybrids provided that the extraction should be done from the seedlings of the same age.
